MtDNA deletion and transcription level in the aged mouse tissues

Abstract

We have investigated the rate of mtDNA deletion and transcription level in mouse heart and smooth muscle tissues of various age (3-,7-,12 and 22months) by PCR and Northern analysis. It was confirmed that mtDNA deletion rate increased with age. Deletion band was detected first from 7 month old mouse and deletion was accumulated thereafter until 22 month. The deletion pattern was different in these tissues; in heart mitochndria, the size of deleted mtDNA was 1.5 kb, 4.1 kb, 5.3 kb, while in smooth mucle, the size of deleted mtDNA was 5.3 kb. Especially, the deletion rate of heart tissues was higher than the rate of smooth muscle¡¯s. The rapid rise in mtDNA mutation suggests that the deleted mtDNA might be preferentially replicated. By Northern analysis experiments, the steady-state concentration of mtDNA undergoes an age-dependent decrease in mouse heart, But the transcription level of smooth muscle was not significantly decreased with age. The transcription level was different in these tissues. This result suggests that the transcription level was affected by mtDNA deletion rate. If mtDNA deletion is increased, impaired transcription will occur. Hence function for survival. Therefore, their effect on energy output increases exponentially with age.
